鰆魚有兩科,帶鰆科 (Gempllidae) 和鯖科 (Scombridae),魚種主要分布於太平洋和印度洋海域,為熱帶及亞熱帶海域之魚種,在台灣大多分布於東部及南部海域。本實驗利用粒線體 DNA (Mitochondrial DNA) 中的 Cytochrome b (Cytb) 基因片段對台灣鰆魚進行物種鑑定,並透過聚合酶連鎖反應-限制酶片段長度多型性 (Polymerase chain reaction-restriction fragment length polymorphism, PCR-RFLP) 建立鰆魚之 PCR-RFLP 圖譜。其結果顯示利用 L14735 和 H15149ad 兩段基因引子可增幅 7 種台灣常見鰆魚之 Cytb gene 之片段序列,長度約為 468-470 bp,另以 BfaI、StuI和 FokI 三種限制酶作片段切位分析,結果顯示利用這三組限制酶可將 7 種台灣常見鰆魚區分,並建立鰆魚之 PCR-RFLP 圖譜。另外也蒐集 11 件台灣土魠魚羹店所販售之土魠魚塊,並以 PCR-RFLP 技術鑑定其魚種。由實驗結果得知,台灣各地土魠魚羹店使用的魚種皆有不同,分別有康氏馬加鰆、棘鰆、巴拉金梭魚、海鱺、黑皮旗魚以及澳洲馬鮫與半線馬鮫的雜交魚種等,其中僅有 1 間店家是以較高價位的康氏馬加鰆 (Scomberomorus commerson) 作為原料來販售。
本研究第二章將透過小鼠生物試驗 (Mouse bioassay) 對鰆魚魚肉進行西卡毒毒性試驗,結果顯示受檢測之台灣鰆魚皆沒有熱帶性海魚毒之中毒風險。再利用高效液相層析儀 (High performance liquid chromatography, HPLC) 分析鰆魚肝臟中維生素 A 含量。結果顯示,其最高含量的維生素 A 在康氏馬加鰆、尖身帶鰆和東方齒鰆肝臟中維生素 A 含量分別為 9,129 ± 18、21,330 ± 340 和 14,409 ± 23 IU/g,其肝臟中維生素 A 總含量分別為 849,727 ± 18、3,350,943 ± 340 和 564,976 ± 23 IU,此三種魚類之維生素 A 總含量皆高於 500,000 IU,具有維生素 A 急性中毒之風險,應避免食用這些魚類的肝臟。為了瞭解鰆魚之魚體大小、肝臟大小和維生素 A 含量之相關性,另外以迴歸分析作圖,得知台灣鰆魚的魚體大小與肝臟大小呈正相關,而魚體大小和維生素 A 含量的相關性因魚種不同而異。
Gempylidae and Scombridae are belong to mackerel fish and mainly inhabit in the sea area of the Pacific and Indian Oceans. It is mostly in the eastern and southern sea area of Taiwan. In this study, the cytochrome b gene fragment of mitochondrial DNA was used to identify the species of Gempylidae and Scombridae in Taiwan. It can be amplify 468-470 bp fragment sequences of Cytb gene on 7 species of Gempylidae and Scombridae fish in Taiwan by using a pair of primers (L14735/H15149ad). Then, Using StuI、BfaI and FokI restriction enzymes, the seven Gempylidae and Scombridae fish can be distinguished by PCR-RFLP method. And the PCR-RFLP map of Gempylidae and Scombridae was established. In addition, eleven fried mackerel meats were collected in Taiwan, and used the PCR-RFLP technology to identify fish species of fried mackerel meat. According to the results, the fish species used in fried mackerel meat in Taiwan are different, including Scomberomorus commerson, Acanthocybium solandri, Sphyraena barracuda, Rachycentron canadum, Makaira nigricans and hybrid fish of Scomberomorus munroi × Scomberomorus semifasciatus. The species of one fried mackerel meat are Scomberomorus commerson in eleven commercial products.
In second part of this study, the ciguatera toxin of Gempylidae and Scombridae fish was detected by mouse bioassay. The result showed there is no risk of ciguatera by eating fish Gempylidae and Scombridae. Further, vitamin A level in the liver of Gempylidae and Scombridae fish by high performance liquid chromatography. The highest level of vitamin A in Scomberomorus commerson, Thyrsitoides marleyi and Sarda orientalis liver was 9,129 ± 18, 21,330 ± 340 and 14,409 ± 23 IU/g, respectively.
The total of vitamin A in Scomberomorus commerson, Thyrsitoides marleyi and Sarda orientalis liver was 849,727 ± 18, 3,350,943 ± 340 and 564,976 ± 23 IU/g, respectively. The total level of vitamin A in the three fish livers was higher than 500,000 IU. There is a risk of vitamin A poisoning by eating the liver of large fish Scomberomorus commerson, Sarda orientalis and Thysitoides marleyi. Through the regression model, the body weight and liver weight of Gempylidae and Scombridae are directly related, and the correlation between body weight and vitamin A varies depending on fish species.
